yatwhatever (matter) there may be relative pronoun (nom. sg. n.), picked up by tat in L2: sets up a whatever…that… ruleजो (वस्तु) — यह सम्बन्धवाचक सर्वनाम है, जिसका उत्तर आगे 'तत्' से मिलता है; अर्थात् 'जो-जो कथावस्तु हो'
syātshould happen to be optative of √as, inside the relative clause: 'whatever may be…'

nāyakasyaunbecoming to the hero — unfitting for the nāyaka's character genitive of relation: names one thing the matter may clash withनायक की (दृष्टि से) — षष्ठी सम्बन्ध; अर्थात् जिस नायक के स्वभाव से वह वस्तु अनुचित पड़ती हो
rasasya vāor to the rasa — clashing with the dramatic mood second genitive; vā = 'or', giving the two things the matter must not offendअथवा रस की (दृष्टि से) — दूसरी षष्ठी, 'वा' से 'या' का भाव; अर्थात् जिन दो के प्रति वह वस्तु अनुचित न हो, वे हैं नायक और रस
yat vastu — whatever matter; nāyakasya rasasya vā — to the hero or to the rasa; anucitaṁ syāt — should happen to be unbecoming.
Rūpa lays down a rule for the playwright. Take any bit of story-material — vastu, a plot element. Ask: does it sit badly with the hero, or with the rasa, the mood the play is building? If it is unbecoming to either — if it does not fit the nāyaka's character or works against the rasa — then it falls under this rule. This line sets up the case; the verdict comes in the next line.
vastu here is the dramatist's raw plot-material — the incidents and situations drawn into the play. anucita ('unbecoming, unfitting') is measured against two things at once: the nāyaka (the hero must stay in character) and the rasa (the reigning mood must not be broken). Both are kept in IAST as technical terms of dramaturgy.

viruddhaṁ tat parityājyam anyathā vā prakalpayet ||
viruddhaṁbeing contrary to it — at odds with the hero or the mood adjective (nom. sg. n.) agreeing with tat, restating anucita as outright oppositionविरुद्ध, प्रतिकूल — 'तत्' का विशेषण; 'अनुचित' को और तीव्र करके 'सर्वथा विरोधी' कह देता है
tatthat matter correlative of yat (nom. sg. n.): 'that very matter' just describedवह (वस्तु) — 'यत्' का प्रतिसम्बन्धी; अर्थात् अभी जिसका वर्णन हुआ वही कथावस्तु
parityājyamis to be dropped altogether gerundive (nom. sg. n.): the primary prescription — discard itसर्वथा त्याग देने योग्य — कृत्य-रूप विधान; यही मुख्य उपाय है कि उसे छोड़ ही दे
anyathāotherwise — in some changed form adverb: names the second option, reworking rather than deletingअन्य प्रकार से, दूसरी तरह से — यह दूसरा विकल्प बताता है, जहाँ मिटाना नहीं बल्कि बदलना है
vāor 'or', offering the alternative to parityājyamअथवा — 'परित्याज्यम्' के प्रति यह विकल्प खड़ा करता है
prakalpayethe (the poet) should recast it causative optative of pra-√kḷp: the dramatist reshapes the matter to fitरच ले, गढ़कर बैठा ले — प्र-√कॢप् की णिजन्त विधिलिङ्; अर्थात् कवि उस वस्तु को अनुकूल बनाकर ढाल ले
viruddhaṁ tat — that contrary matter; parityājyam — is to be dropped; anyathā vā — or else in a changed form; prakalpayet — he should recast it.
Now the verdict. That contrary matter — viruddha, standing against the hero or the mood — has two fates, and the poet chooses. First option: parityājya, throw it out completely. Second option: keep it, but anyathā prakalpayet — recast it in some altered form so that it no longer clashes. So the rule is: incongruous story-material is either cut, or rewritten to fit. The playwright is free to modify even received material for the sake of the hero and the rasa.
anyathā prakalpayet ('recast it otherwise') names the standard dramaturgical license: the poet may alter received plot-material — even from itihāsa or an earlier play — so that it agrees with the nāyaka and the rasa. prakalpayet is the causative of pra-√kḷp, 'to fashion, arrange, contrive', kept as the working verb for a poet reshaping his matter.

yat vastu nāyakasya rasasya vā anucitaṁ viruddhaṁ syāt, tat parityājyam, anyathā vā prakalpayet।
Whatever bit of plot-material happens to be unbecoming — at odds with the hero's character or with the reigning rasa — that material is to be dropped altogether; or else the poet should recast it in some altered form so that it no longer clashes.
Rūpa's defining kārikā laying down the rule for incongruous vastu: story-material that offends the nāyaka or the rasa must be either cut or reworked to fit.
