Rūpa is defining janāntika, one of the stage-asides. The speaker raises the tripatākā hand — three fingers up — as the signal for a private word. With that gesture he screens off the other characters, as though they cannot hear. What is spoken this way, aside and out of the others' earshot, is a bit of talk — a kathā. That word carries over into the next line.
त्रिपताका नामक हस्तमुद्रा से अन्य पात्रों को ओट करके, अर्थात् मानो वे सुन न सकें ऐसे भाव से, बीच में जो बात कही जाती है। यहाँ लक्षण का पूर्वार्ध है; क्रिया और संज्ञा आगे L2 में पूरी होती है।
janāntika is a technical nāṭya term: an aside spoken on stage between two characters that the other characters present are conventionally treated as not hearing. The tripatākā-kara ('three-fingered flag' hand) is the codified gesture that marks the speech as private — the actor raises it toward the others to signal 'this is not for you.' It contrasts with sva-gata (an aside spoken to oneself) and apavārita (a secret told to one person).

Now the definition closes. The speech that is carried on this way — passed back and forth between just the two characters, with the others screened off — that is what is called janāntika. So janāntika needs three things together: the tripatākā gesture, the shutting-out of the others, and an exchange between exactly two people. Meet all three and the aside earns the name.
जो बात परस्पर दो पात्रों के बीच की जाती है, वह जनान्तिक कहलाती है। इस प्रकार त्रिपताका मुद्रा से औरों को ओट करके ठीक दो जनों में जो वार्ता चलती है, उसी को नाट्य में जनान्तिक नाम से जाना जाता है।
