hāsaḥ — laughter broke out; vandi-jana-uktitaḥ — prompted by what the court-bards said; rājendratā-prasaṅgena — coming up in the course of [talk of] emperorship.
Laughter broke out at what the court-bards said. It came up in the course of their hailing [him] as emperor, king of kings.
These two lines sit under the garbha-sandhi ('womb / germ juncture'), defined just above in kārikā 78: the bīja — the plot-seed once seen and then lost from view (hrāsa) — is sought out anew (gaveṣaṇa). Rūpa is illustrating that re-emergence, giving instances of how the seed resurfaces.

prasena-anveṣaṇātgrowing out of the search for Prasena ablative of source — the earlier hunt is what a fresh search grows out of; the compound = 'the searching for Prasena'प्रसेन की खोज से — इसी पहली खोज से नयी खोज उपजी (अपादान)
prasena-anveṣaṇāt — out of the search for Prasena; punaḥ — once more; anveṣaṇaṁ — a fresh searching; jātaṁ — arose.
Out of the search for Prasena, a fresh search arose. The seeking starts up all over again.
prasena-anveṣaṇa — the hunt for Prasena, from the Syamantaka-jewel episode of Kṛṣṇa's līlā: Prasena, brother of Satrājit, rode out wearing the gem and was killed by a lion, and the search for him opened straight into a further search — for the jewel, then for Jāmbavān. That is exactly the garbha pattern: the plot-seed, lost from view, resurfaces through renewed seeking (punar anveṣaṇa).
rājendratā-prasaṅgena vandi-jana-uktitaḥ hāsaḥ [jātaḥ]; prasena-anveṣaṇāt punaḥ anveṣaṇaṁ jātam.
prasaṅgena rājendratā (उठ आने पर) vandi-jana-uktitaḥ hāsaḥ (jātaḥ); prasena-anveṣaṇāt punaḥ anveṣaṇaṁ jātam।
As talk turned, in the course of it, to his being emperor, king of kings, laughter broke out at what the court-bards said in his praise; and out of the search for Prasena a fresh search arose all over again.
Rūpa's illustrative kārikā for the garbha-sandhi (kārikā 78): two instances of its signature pattern — the plot-seed, once lost from view, sought out anew (punar anveṣaṇa).
