The verse — decoded
L1definition
phale pradhāne bījasya prasaṅgoktaiḥ phalāntaraiḥ |
phale pradhānewhen the main fruit — the plot's central goal — locative absolute, head clause; completed by vicchinne in L2जब प्रधान फल — बीज से मिलने वाला मुख्य फल — (यह वाक्य आगे 'विच्छिन्ने' से पूरा होता है)
bījasyaof the bīja — the seed sown at the play's start genitive dependent on phale: the fruit that the seed was to yieldबीज का — अर्थात् जो फल बीज से उत्पन्न होना था
prasaṅgoktaiḥby [matters] brought up only in passing, as the occasion arose instrumental of means, pairs with phalāntaraiḥप्रसंगवश आ पड़े — बीच में जो प्रसंगवश कहे गए, उनके द्वारा
phalāntaraiḥby other, side results — a second line of outcome instrumental of means; the agent of the interruptionअन्य फलों के द्वारा — जो मुख्य फल में विच्छेद डालते हैं
bījasya pradhāne phale — the main fruit of the seed; prasaṅgoktaiḥ phalāntaraiḥ — [is interrupted] by other, secondary results brought up in passing.
Now the second plot-element is being defined — the bindu, the 'drop'. Picture the seed's main fruit: the central goal that was set moving at the play's opening. As the action unfolds, other side-results come up in passing and, for a stretch, the main goal is pushed aside.
This is the received dramaturgical definition of the bindu, the 'drop' — the second of the five arthaprakṛtis, the plot-driving elements (bīja the seed, bindu the drop, patākā the banner-subplot, prakarī the episode, kārya the final aim). The seed (bīja) is the small germ of the main theme sown at the start; the bindu is what carries that theme across an interruption. The lead-in just before this verse rounds off the previous kārikā's bīja by pointing to the Lalita-Mādhava, where the matured love (anurāga) of Rādhā and Mādhava, set in the mukha-sandhi of acts one and two, is the seed; the illustration of the bindu itself follows in the next kārikā's lead-in (the garbhāṅka of acts three and four).
L2definition

vicchinnewhen it is cut off, broken away for a while locative absolute completing phale pradhāne of L1: 'when the main fruit is interrupted'जब वह विच्छिन्न (बीच में टूट) जाए — यही 'phale pradhāne' को पूरा करता है: 'जब मुख्य फल में विच्छेद पड़ जाए'

vicchinne — when [the main fruit] is cut off; yat — that which; aviccheda-kāraṇaṁ — is the cause of its non-interruption; binduḥ ucyate — is called the bindu.
When the main goal is thus broken off, whatever holds it together — whatever carries the main theme through the gap so it can resume — that is called the bindu. The name means a 'drop': as a single drop of oil spreads and holds across a surface of water, the bindu is the thin thread that keeps the main plot alive while the side-matters run their course.
aviccheda-kāraṇa — the 'cause of non-interruption' — is the whole point of the term: the bindu is not the interruption but the remedy for it, the continuity-thread. The verse closes the definition with 'bindur ucyate', fixing the technical name; it comes into Rūpa's treatise from the shared nāṭya-śāstra tradition, and the Lalita-Mādhava illustration of it (the play-within-a-play, garbhāṅka, of acts three and four) is supplied in the following kārikā.
bījasya pradhāne phale prasaṅga-uktaiḥ phala-antaraiḥ vicchinne (sati) yat aviccheda-kāraṇam (bhavati), (tat) binduḥ ucyate.

When the seed's main fruit — the central goal that was set in motion at the play's opening — is broken off for a while by other, side results that come up only in passing as the occasion arises, then whatever becomes the cause of its non-interruption — whatever carries the main theme across the gap so that it can resume — is called the bindu, the 'drop'.
The received definition (lakṣaṇa) of the bindu, the 'drop' — the second of the five arthaprakṛtis (plot-driving elements: bīja, bindu, patākā, prakarī, kārya); the lead-in sets up its Lalita-Mādhava illustration in the garbhāṅka of acts three and four.
