L1definition
ekaikasyās tv avasthāyāḥ prakṛtyā caikayaikayā |
ekaikasyāḥof each single stage, taken one at a time gen. sg., agreeing with avasthāyāḥ; eka-eka = 'one by one,' distributiveप्रत्येक एक-एक (अवस्था का) — 'एक-एक' कहकर एक-को-एक का मेल दर्शाया गया है
tunow — turning to define the juncture itselfकिन्तु (अब लक्षण कहा जाता है)
avasthāyāḥof a stage (avasthā) — one of the phases the plot's action passes through gen. sg.; this is the term being paired offअवस्था का — जिसे जोड़ा जा रहा है वह पहला अंग
prakṛtyāwith a plot-element (prakṛti) — the thing joined to that stage instr. of the second partner in the pairingएक प्रकृति के साथ — मेल का दूसरा अंग
caandऔर
ekayā ekayāwith just one at a time — one element matched to one stage instr., reduplicated to force the one-to-one pairing (not many elements to one stage)एक-एक के साथ — दोहराकर यह जताया कि एक अवस्था का एक ही प्रकृति से मेल हो (अनेक का एक से नहीं)
ekaikasyāḥ avasthāyāḥ — of each single stage; ekayā ekayā prakṛtyā ca — and with each single plot-element; [the joining of these, defined in L2].
Now Rūpa defines the sandhi, the juncture. Take the stages the plot's action moves through (avasthā), and take the plot-elements (prakṛti). Pair them off one by one — one stage to one element at a time. The doubled 'one … one' is the point: it is a strict one-to-one match, not a crowd of elements piled onto a single stage.
This uses two technical sets from the nāṭya-śāstra. The avasthās are the five stages the action passes through toward its goal — ārambha (beginning), yatna (effort), prāptyāśā (hope of attainment), niyatāpti (certainty of attainment), and phalāgama (arrival of the fruit). The prakṛtis (arthaprakṛtis, the 'plot-elements' or means) are five as well — bīja, bindu, patākā, prakarī, and kārya — which the very next kārikā (5.2) names in order. Because each set has five members, pairing them one-to-one yields five junctures.

[ekaikasyāḥ avasthāyāḥ ekayā ekayā prakṛtyā ca] yogaḥ — the joining of each stage with each single element; sandhiḥ iti — is 'a sandhi'; nāṭya-vidyā-viśāradaiḥ jñeyaḥ — so those skilled in dramaturgy should know it.
That joining — one stage locked with one plot-element — is what the experts in dramaturgy are to recognize as a sandhi, a juncture. So the whole definition reads: the union of each single stage with each single element is called a sandhi. Since there are five stages and five elements, this yields the five junctures of a play.
The five sandhis that result from these pairings are mukha, pratimukha, garbha, avamarśa (also vimarśa), and nirvahaṇa — the standard five junctures of a Sanskrit drama, each binding one phase of the plot to the next. jñeyaḥ is a gerundive of prescription: this is not a stray usage but the settled term the trained dramatist is expected to know.
ekaikasyāḥ tu avasthāyāḥ ekayā ekayā prakṛtyā ca yogaḥ sandhiḥ iti nāṭya-vidyā-viśāradaiḥ jñeyaḥ.
ekaikasyāḥ tu avasthāyāḥ ekayā ekayā ca prakṛtyā (यः) yogaḥ (भवति), (सः) sandhiḥ iti nāṭya-vidyā-viśāradaiḥ jñeyaḥ।
The joining of each single stage of the plot's action (avasthā) with each single plot-element (prakṛti), paired strictly one to one, is what those skilled in the science of drama are to recognize as a sandhi, a juncture. Since the stages are five and the plot-elements five, this one-to-one matching yields the five junctures of a play.
Rūpa's defining kārikā (lakṣaṇa) of the sandhi: it defines a juncture as the one-to-one pairing of an avasthā with a prakṛti, opening the vibhāga on 'sandhiḥ'.
